چکیده

Regeneration of dental pulp after pulpectomy was accomplished recently by autologous transplantation of dental pulp stem/ progenitor cells into the root canal. The identical patterns of qualitative and quantitative protein and mRNA expression in the regenerated pulp and normal pulp demonstrated complete pulp regeneration. The lack of tissue specific markers in the dental pulp is a major challenge in dental regenerative medicine. In order to identify specific markers in dental pulp we undertook a comparison of the gene expression profile of the dental pulp with that of periodontal ligament and gingiva. This systematic investigation identified thyrotropinreleasing hormone (TRH)-degrading enzyme (DE) as a marker of dental pulp. Expression of TRH-DE mRNA in human dental pulp was higher than that in any other tissue except brain as analyzed by real time RT-PCR. Induction of neural cells enhanced the expression of TRH-DE mRNA in dental pulp stem/progenitor cells (CD105+ and CD31side population (SP) cells) in vitro. Immunohistochemical and in situ hybridization analyses demonstrated that TRH-DE in the neuronal processes in dental pulp. In canine pulp cells, TRH down-regulated TRH-DE mRNA expression, while neuropeptide Y up-regulated it, suggesting that TRH-DE has functional role in the neuropeptide signaling in dental pulp tissue. It is noteworthy that TRH-DE mRNA was expressed in the regenerated pulp 28 days after transplantation of CD31SP cells into root canals after pulpectomy. These results demonstrate the utility of TRH-DE as a novel dental pulp biomarker during regeneration of pulp. *Corresponding author: Misako Nakashima, Department of Dental Regenerative Medicine, Center of Advanced Medicine for Dental and Oral Diseases, National Center for Geriatrics and Gerontology, Research Institute, 35 Gengo, Morioka, Obu, Aichi 474-8511, Japan, Tel: 81 562 44 5651 (ext 5065); Fax: 81 562 46 8684; E-mail: [email protected] Received October 20, 2011; Accepted December 12, 2011; Published January 14, 2012 Citation: Yamamoto T, Murakami M, Ishizaka R, Iohara K, Kurita K, et al. Purification and characterization of the thyrotropin-releasing hormone (TRH)-degrading serum enzyme and its identification as a product of liver origin.

Previous biochemical studies have indicated that the membrane-bound thyrotropin-releasing hormone (TRH)-degrading enzyme (TRH-DE) from brain and liver and the serum TRH-DE are derived from the same gene. These studies also suggested that the serum enzyme is of liver origin. The present study was undertaken to verify these hypotheses. In different species, a close relationship between the activi...

Identification of the thyrotropin-releasing-hormone-degrading

A time-dependent inhibition of the thyrotropin-releasing-hormone (TRH)-degrading ectoenzyme (EC 3.4.19.-) from rat brain by the metal-complexing agents imidazole, NaCN, EDTA and 1,10-phenanthroline could be demonstrated. In contrast, the inhibition by the non-chelating analogues 4,7and 1,7-phenanthroline was not time-dependent. At a concentration of 100 ,M EDTA the enzymic activity decreased by...

Characterization of a pyroglutamate aminopeptidase from rat serum that degrades thyrotropin-releasing hormone.

An enzyme which degrades thyrotropin-releasing hormone (TRH) into pyroglutamic acid and histidylprolineamide has been partially purified from rat serum. The partially purified enzyme was separated from other serum enzymes that degrade TRH. The products formed by the purified enzyme were established with [L-proline-2,3-“HIand [L-<GZu-( U)-14C]TRH using thin layer chromatography arid electrophore...

Lower levels of thyrotropin-releasing hormone-degrading activity in human cord and in maternal sera than in the serum of euthyroid, nonpregnant adults.

Thyrotropin-releasing hormone (TRH)-degrading activity was investigated in human cord, maternal, and euthyroid adult sera by measuring (a) the rate of disappearance of TRH and (b) the rate of formation of degradation products. The rate of TRH degradation in cord and maternal sera was 25-33% of that in euthyroid adult serum. Concomitantly, in cord and maternal sera, the rate of formation of prol...

G PROTEIN-COUPLED RECEPTOR SIGNALLING IN NEUROENDOCRINE SYSTEMS Thyrotropin-releasing hormone receptors – similarities and differences

Thyrotropin-releasing hormone (TRH) initiates its effects by interacting with cell-surface membrane receptors. Two G protein-coupled receptors for TRH, TRH receptor type 1 (TRH-R1) and TRH receptor type 2 (TRH-R2), have been cloned from mammals. In this review, we compare TRH-R1 and TRH-R2 with regard to their tissue distribution, binding affinities for TRH and TRH analogs, basal and activated ...
